Induction of Programmed Cell Death in Human Breast Cancer Cells by an Unsymmetrically Alkylated Polyamine Analogue1

چکیده
The need for antineoplastic compounds with novel mechanisms of ac tion is great. One such agent is the recently synthesized polyamine analogue /V'-ethyl-A'n-((cyclopropyl)methyl)-4,8-diazaundecane (CPENSpm). Expo sure of hormone-dependent and -independent human breast cancer cells to 0.1-10 UM CPENSpm led to both growth inhibition and induction of pro grammed cell death. Fragmentation of DNA to high molecular weight frag ments and oligonucleosomal-sized fragments, both characteristic of pro grammed cell death, was determined to be time and concentration dependent Depletion of natural polyamine pools and accumulation of the analogue was also demonstrated. These data provide the first evidence that a polyamine analogue induces programmed cell death.

The need for antineoplastic compounds with novel mechanisms of action is great. One such agent is the recently synthesized polyamine analogue N1-ethyl-N11-((cyclopropyl)methyl)-4,8-diazaundecane (CPENSpm). Exposure of hormone-dependent and -independent human breast cancer cells to 0.1-10 microM CPENSpm led to both growth inhibition and induction of programmed cell death. Fragmentation of DNA to...
متن کاملPolyamine analogue induction of programmed cell death in human lung tumor cells.
The naturally occurring polyamines putrescine, spermidine, and spermine are required for cell growth. Based on this requirement, several polyamine analogues that interfere with polyamine function and metabolism have been synthesized as antineoplastic agents. The symmetrically substituted N1,N12-bis(ethyl)spermine (BESpm), and unsymmetrically substituted N1-ethyl-N11-[(cyclopropyl)methyl]-4, 8-d...
متن کاملInduction of Apoptosis and Non-Apoptosis in Human Breast Cancer Cell Line (MCF-7) by Cisplatin and Caffeine
Background: Molecular targeted therapy by different cell death inducers are recently considered in cancer therapy. The aim of this study was to compare the effect of cisplatin and inositol trisphosphate kinase inhibitor (caffeine) on human breast cancer cell line (MCF-7). The pattern of cell death in MCF-7 cells following the exposure to cisplatin and caffeine in individual and combination form...
متن کاملG1 Phase Arrest and Apoptosis Induction in Human Thyroid Cancer Cell Line Thr.C1.PI33 by 3-Hydrogenkwadaphnin Isolated from Dendrostellera lessertii
Dendrostellera lessertii (Thymelaeaceae) is a toxic plant that grows in parts of Iran. The anti-proliferative properties of its crude methanol extract and one of its active components, 3-hydrogenkwadaphnin (3-HK), have been established using several cancer cell lines. Methods: In a further attempt to determine the mode of action, two groups of synchronously growing cells were treated with a sin...
متن کاملUnsymmetrically substituted polyamine analogue induces caspase-independent programmed cell death in Bcl-2-overexpressing cells.
The polyamine analogue, N1-ethyl-N11-[(cycloheptyl)methyl]-4,8-diazaundecane (CHENSpm)-induced programmed cell death in NCI H157 cells is accompanied by cytochrome c release, the loss of mitochondrial membrane potential, activation of caspase-3, caspase-mediated poly(ADP-ribose) polymerase cleavage, G2-M arrest, and DNA and nuclear fragmentation. Overexpression of Bcl-2 completely inhibits CHEN...
